As you are saying that you are not able to run Windows defender in your windows vista powered PC so, you should restore your system once then check the issue that should be resolved after performing process of system restoring in Windows vista PC. Follow the procedure and make a note that you are not having any media in your DVD/CD writer:

Step 1: Restart PC and for Single OS user, press & hold f8 as the computer restarts. Before the Windows logo appears, press and hold F8. If more than one operating system using then highlight OS by arrow keys and select Safe Mode then press Enter.
Step 2: Now, log on PC with administrator account
Step 3: Click on start button and navigate to All Programs
Step 4: Select Accessories and then system tools
Step 5: Click on the System Restore
Step 6: Screen for System Restore wizard will be appeared and follow steps to return previous restore point

    Hi.
check for the status of “Windows Defender” service and ensure that it’s started and running.
    a. Click on Start.
    b. Type “services.msc” in the Start Search box and hit Enter on the keyboard.
    c. Double-click on Windows Defender.
    d. Under Service status in the General tab, click Start to start the service.
    e. From the startup type dropdown menu select Manual.
    f. Click on Apply and OK.

    Thank you.
